- First non-standalone Pop'n Music game to include Animelo songs.
- First game in the series to include the singing of ALPHA, who would sing many, many songs with Osamu Migitera (Des-ROW) over the years.
- First appearances of Guitar Freaks mainstays Hideyuki Ono and Ueko Harumi (Jimmy Weckl) in the series.
- First appearance of Tomoyuki Uchida (Mr.T) in the series.
- Mimi and Nyami are drawn in a slight more anime style, but otherwise don't look much different from Pop'n Music 5.
